Jefferson played college basketball for the Arizona Wildcats. He was drafted with the 13th overall pick in the first round of the 2001 NBA draft. Jefferson was named to the NBA All-Rookie Second Team following his first NBA season. He played seven seasons for the New Jersey Nets (later known as the Brooklyn Nets), who reached the NBA Finals in each of his first two seasons. Jefferson later played for the Milwaukee Bucks, the San Antonio Spurs, the Golden State Warriors, the Utah Jazz, the Dallas Mavericks, the Cleveland Cavaliers, and the Denver Nuggets. He won an NBA championship as a member of the Cavaliers in 2016. Jefferson was also a member of the United States national team that won a bronze medal in the 2004 Summer Olympics.
Jefferson retired from the NBA in 2018, becoming a basketball analyst. He joined the staff of ESPN in 2019.
