Kuko sąsiauris (angl. Cook Strait, maor. Te Moana-o-Raukawa) – sąsiauris, skiriantis pagrindines Naujosios Zelandijos salas – Šiaurės ir Pietų. Šiaurinėje pusėje pastatyta sostinė Velingtonas, o pietinėje – Marlboro Saundsas ir Klaudi Bėjus. Vidutinis sąsiaurio gylis apie 128 metrų. Ilgis 107 km, plotis 22–91 km.[1] Yra seklių įlankų. Daug mažų salų ir uolų. Potvyniai kas pusę paros; aukštis apie 4 m.
Dvi didelės įlankos Auksinė (Golden Bay) ir Tasmano giliai įsirėžusios į Pietų salą, į vakarus nuo sąsiaurio. Šiaurės saloje, į vakarus nuo sąsiaurio, tęsiasi ilga Kapiti pakrantė. Siauriausia sąsiaurio vieta yra ten, kur susieina Teravičio pusiasalis (Šiaures saloje) ir Aradavos sala (šalia Marlboro Saundso).
Sąsiauriu nuolat kursuoja keltas iš Velingtono į Marlboro Saundsą. Kuko sąsiauryje dažni audringi vėjai ir didelės bangos.
Sąsiauris taip buvo pavadintas todėl, kad 1769 metais Džeimsas Kukas praplaukė šį sąsiaurį pirmasis iš europiečių. Tiesa, jau anksčiau (1642 m.) jį pasiekė Abelis Tasmanas, tačiau palaikė įlanka.[2]
