El Torneo de la División I de Baloncesto Masculino de la NCAA de 2010 se celebró para determinar el Campeón de la División I de la NCAA. Fueron 65 los equipos que disputaron la fase final, jugándose la Final Four en el Lucas Oil Stadium de Indianápolis, Indiana.[1]

Los ganadores fueron el equipo de la Universidad de Duke, que lograban su cuarto campeonato, el anterior en 2001, derrotando en la final a la Universidad Butler, en su primera aparición en una final. Kyle Singler, de los Blue Devils, fue considerado Mejor Jugador del Torneo.
Fue la primera vez en 25 años que se enfrentaban en la final dos instituciones privadas, desde que lo hicieran Villanova y Georgetown en 1985, y la quinta vez que sucedía a lo largo de la historia del campeonato.
