Mistra garsėja kaip buvusi Bizantijai priklausiusios Morėjos despotystės (XIV–XV a.), apėmusios Peloponeso pusiasalį, sostinė. Čia išlikęs gausus viduramžių architektūros paveldas: tvirtovė, Brontocho, Periblepto vienuolynų liekanos, veikiantis Pantanasos vienuolynas, keletas bažnyčių, vartai, įvairūs rūmai, gyvenamieji ir ūkiniai pastatai.
Senovėje šioje vietovėje gyveno slavų gentis milingai. XIII a. vid. Achėjos kunigaikštis Viljamas Vilarduenas Mistroje pastatė tvirtovę. 1259 m. ji užkariauta Nikėjos imperijos (Bizanijos). 1262 m. Mistra tapo Morėjos despotystės sostine. Mieste suklestėjo menai, filosofija (čia mokė neoplatonikas Plitonas), statyti vienuolynai, išpuošti freskomis. 1460 m. Mistrą užėmė Osmanai, jos reikšmė sumenko. XIX a. dauguma Mistros gyventojų persikėlė į naujai įkurtą Spartą.
1989 m. Mistra paskelbta UNESCO pasaulio paveldo objektu.
