Shirley MacLean Beaty var fødd i Richmond i Virginia. Broren Warren Beatty blei også seinare skodespelar.

MacLaine debuterte i Hitchcock-filmen The Trouble with Harry (1955) som ho vann ein Golden Globe-pris som årets nykomling for. I 1956 spelte ho i Hot Spell, Around the World in Eighty Days og Some Came Running. Den sistnemnde fekk ho ein Golden Globe-nominasjon og sin første Oscar-nominasjon for. Etter fleire nominasjonar for andre filmar vann ho sin første oscarpris i 1983 for Terms of Endearment.
