Det ble grunnlagt i 1438 av kong Henrik VI, Henry Chichele som var fellow ved New College og erkebiskopen av Canterbury, til minne om de falne under hundreårskrigen. Den er fremfor alt en forskningsinstitusjon.
En unik særegenhet er at dette college ikke har noen studenter uten avslutningsrksamen; alle er fellows, det vil si utdannede akademikere.
Medlemskapet rekrutteres blant annet ved at men hvert år inviterer University of Oxfords beste absolventer til å skrive en eksamen. På grunnlag av disse opptakseksamener velges to kandidater til nye fellows. Et fellowship ved All Souls College er en av Englands ypperste akademiske utmerkelser.
I selve kollegiet omgåes dermed en engere krets av unge forskere med allerede akademisk etablerte størrelser i tillegg til mennesker som har gjort karriere i politikk, næringsliv eller medier.
Det stilfulle fasadebygget er forblitt nærmest uendret siden det stod ferdig i 1443. Av særlig interesse er hovedbygget fra 1733, Great Quadrangle, særlig dobbelttårnene tegnet av Nicholas Hawksmoor, og soluret fra 1658 av Christopher Wren.
