Universitetet i Alcalá (spansk: Universidad de Alcalá de Henares) var et 1500- og 1600-tallets mest betydelige universiteter. Det ble grunnlagt i 1499 i Alcalá de Henares og flyttet til Madrid i 1836. I dag er dets etterfølgeruniversitet, Universidad Complutense de Madrid[1], Spanias største universitet. Det historiske universitetets bygningeri Alcalá ble i 1998 med byen rundt erklært til kulturarv av UNESCO.

I 1977 ble det i Alcalá grunnlagt et nytt offentlig universitet ved navn Universidad de Alcalá, som ved siden av tidligere ordenskonventer og nye bygninger også benytter bygmninger fra det gamle universitetet.[2]
Allerede før den pavelige grunnleggelse i 1499 ved Alexander VI etter petisjon fra en elev av Gonzales Jiménez de Cisneros fantes det i Alcala siden 1293 en av kong Sancho IV anerkjent fransiskansk læreanstalt med et Studium generale.
Universitetet innebar at Alcalá de Henares ble fullstendig ombygd etter en ny plan; det skulle være en modell på en Civitas Dei (Guds by), en idealby som skulle inspirere de spanske misjonærer rundt om i verden. Her skulle man se i arkitektur og byplan en slik idealby.
AUniversitetes oppgave skulle etter de Cisneros' konstitisjoner være å utdanne kompetente klerikere, som skulle kunne motvirker reformasjonens folk. Derutover skulle det utdannes sakkyndige riksembedsmanne for de forskjellige spanske kolonier.
Mot slutten av 1700-tallet førte studiereformer til at universitet tapte sin lederstilling i Europa, og at det i 1836 ble flyttet til Madrid. Der het det til 1951 Universidad Central, idet det var dey eneste spanske universitet som gav doktorgrader. Med dette monopolets fall fikk ned navnet Universidad Complutense de Madrid.
